Posidinemab (JNJ-63733657) is a humanized IgG1 monoclonal antibody designed to target the microtubule-associated protein tau (MAPT), specifically binding to pathological tau species. Developed by Janssen Research & Development (a subsidiary of Johnson & Johnson), it is being investigated for the treatment of early Alzheimer's disease. The mechanism involves binding to pathological tau aggregates to prevent their trans-neuronal spread (the seeding hypothesis), thereby potentially slowing cognitive and functional decline. It is currently being evaluated in Phase 2 clinical trials, including the Autonomy study, which focuses on patients with early-stage Alzheimer's disease.
