Posoleucel is an investigational allogeneic (off-the-shelf) multi-virus specific T cell therapy designed to prevent or treat infections caused by six major viruses in immunocompromised patients—adenovirus (AdV), BK virus (BKV), cytomegalovirus (CMV), Epstein-Barr virus (EBV), human herpesvirus 6 (HHV-6), and JC virus. It consists of HLA-matched donor-derived memory CD4+ and CD8+ T cells with specificity for these target viruses. Posoleucel works by providing adoptive transfer of functional antiviral T cells to restore immunity in patients with severe deficits in cellular immune function following solid organ or hematopoietic stem cell transplantation. The product is enriched for central and effector memory T cells capable of mounting a broad Th1-polarized antiviral response upon exposure to the target pathogens[2][3][5]. Developed originally at Baylor College of Medicine and advanced by AlloVir/Kalaris Therapeutics, posoleucel has received multiple regulatory designations including FDA Regenerative Medicine Advanced Therapy (RMAT) status and Orphan Drug Designation for several indications[6][9][10].
