Povetacicept is a recombinant fusion protein engineered as a dual antagonist of B cell-activating factor (BAFF, also known as TNFSF13B) and a proliferation-inducing ligand (APRIL, also known as TNFSF13). It is designed to inhibit both BAFF and APRIL signaling by binding to these cytokines with high affinity via an engineered transmembrane activator and CAML interactor (TACI) domain fused to an Fc fragment. This dual inhibition leads to reduced survival and differentiation of B cells and plasma cells, resulting in decreased production of immunoglobulins implicated in autoimmune diseases. Povetacicept has demonstrated potent pharmacodynamic effects in preclinical models and early clinical studies, including reductions in antibody-secreting cells and disease-related biomarkers such as galactose-deficient IgA1. The drug is being developed primarily for autoimmune kidney diseases like IgA nephropathy but is also under investigation for other B-cell-mediated autoimmune disorders[2][5][9].
