Poxatinib (also known as HS-25) is a third-generation small-molecule tyrosine kinase inhibitor (TKI) developed by Hisun Pharmaceutical. It is a structural analog and isostere of ponatinib, utilizing a pyrazolo[1,5-a]pyrimidine core instead of the imidazo[1,2-b]pyridazine core found in ponatinib. Poxatinib is designed to inhibit the BCR-ABL fusion protein, including the T315I gatekeeper mutation, which confers resistance to first- and second-generation TKIs. It is primarily developed for the treatment of chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL). Clinical development in China includes Phase 1/2 trials in patients with resistant or intolerant leukemia and bioequivalence studies comparing poxatinib phosphate to the reference drug ponatinib (Iclusig).
