Poziotinib + paclitaxel + trastuzumab is an investigational combination therapy composed of three agents: poziotinib, a potent irreversible pan-HER tyrosine kinase inhibitor that targets EGFR, HER2 (ERBB2), and HER4 (ERBB4); paclitaxel, a microtubule stabilizing antineoplastic agent; and trastuzumab, a monoclonal antibody targeting the HER2 extracellular domain. This combination is primarily studied for the treatment of HER2-positive advanced gastric cancer, with poziotinib enhancing the efficacy of paclitaxel and trastuzumab by irreversibly inhibiting HER-family signaling pathways and overcoming resistance mechanisms via inhibition of ABCB1-mediated drug efflux. Paclitaxel disrupts microtubule dynamics and mitosis, while trastuzumab inhibits HER2 signaling, leading to growth arrest and apoptosis in HER2-overexpressing tumors.
