**pp-PA83** is a plant-produced, precursor form of the 83 kDa protective antigen (PA83) from *Bacillus anthracis*, engineered in *Nicotiana benthamiana* for use in anthrax subunit vaccines. It consists of a pro-protein (pp) version that requires processing to mature PA63 for full activity, but glycosylation in plants impairs its biological function and stability. Co-expression with peptide-N-glycosidase F enables *in vivo* deglycosylation to produce the active, non-glycosylated variant pp-dPA83, which exhibits comparable *in vitro* cell-binding and pore-forming activity to prokaryotic rPA83, induces superior toxin-neutralizing antibodies in mice, and demonstrates enhanced thermostability. Developed as a low-cost, scalable vaccine antigen candidate, it adsorbs effectively to Alhydrogel adjuvant via electrostatic interactions without major structural disruption, supporting its potential for mass immunization against anthrax bioterrorism.[3][6]
