Overview

**PP353** is an investigational, locally administered formulation of micronized **linezolid** developed by Persica Pharmaceuticals for chronic low back pain associated with Modic type 1 vertebral endplate changes. It is prepared as a 50 mg/mL linezolid suspension in a thermosensitive, radio-opaque vehicle containing poloxamer 407 and iohexol; the product is injected percutaneously into the nucleus pulposus, where it forms a local depot at body temperature. Linezolid is an oxazolidinone antibacterial that binds bacterial 23S ribosomal RNA in the 50S ribosomal subunit, blocking formation of the 70S translation-initiation complex and thereby inhibiting bacterial protein synthesis. PP353 is intended to provide sustained, high local antibacterial exposure in a potentially infected intervertebral disc while limiting systemic exposure.
