PP4 siRNA is a small interfering RNA (siRNA) designed to silence the expression of Protein Phosphatase 4 (PP4), specifically targeting the catalytic subunit PPP4C. PP4 is a serine/threonine phosphatase that plays a critical role in cell cycle progression, centrosome maturation, and DNA damage repair. Research presented at the AACR 2015 conference highlighted PP4 as a potential therapeutic target in BRAF wild-type melanoma, where it is often overexpressed and associated with disease progression and resistance to BRAF and MEK inhibitors. In preclinical studies using human melanoma cell lines, treatment with PP4 siRNA resulted in significant reduction of PP4 protein levels, leading to inhibited cell proliferation, cell cycle arrest, and the induction of apoptosis. This modality represents a targeted approach to treating metastatic melanoma through a BRAF- and MEK-independent pathway.
