pp65 RNA-LPs is an investigational mRNA-based cancer vaccine consisting of messenger RNA encoding the Cytomegalovirus (CMV) phosphoprotein 65 (pp65) encapsulated within lipid particles (LPs). Developed by researchers at the University of Florida, this immunotherapy is designed to treat malignancies that express CMV antigens, such as pediatric high-grade gliomas (pHGG). The mechanism involves the intravenous administration of the RNA-LPs, which primarily localize to the lungs and lymphoid organs to transfect antigen-presenting cells (APCs). These activated APCs then induce a robust, antigen-specific T-cell response and remodel the systemic and intratumoral immune environment to overcome immunosuppression. It is being studied both as a monotherapy and in combination with other tumor-specific mRNA-LPs in the RNA PRIME clinical trial.
