PP6c siRNA is a small interfering RNA (siRNA) designed to silence the expression of the catalytic subunit of protein phosphatase 6 (PP6c, encoded by the PPP6C gene). PP6 is a serine/threonine protein phosphatase that plays a critical role in the DNA damage response by regulating the activation of DNA-dependent protein kinase (DNA-PK). Research, including studies from Shanghai Jiao Tong University, has demonstrated that PP6c is overexpressed in glioblastoma (GBM) and correlates with poor patient survival and radioresistance. By knocking down PP6c, this siRNA inhibits DNA-PK activation and reduces the DNA-repair capacity of tumor cells, thereby sensitizing them to radiotherapy. In preclinical xenograft models, PP6c knockdown has been shown to increase radiosensitivity and prolong survival, positioning it as a potential therapeutic strategy for enhancing the efficacy of radiation in aggressive cancers.
