Overview

PPC-001 (also known as PRP) is an investigational, non-cytotoxic proenzyme-based combination therapy being developed by Propanc Biopharma for the treatment of advanced solid tumors, including pancreatic, ovarian, and colorectal cancers. It consists of two naturally derived pancreatic proenzymes (zymogens)—trypsinogen and chymotrypsinogen A—in a synergistic fixed 1:6 weight-to-weight ratio. Administered via weekly intravenous infusion, these inactive precursors are activated in vivo into trypsin and chymotrypsin, which act as agonists on proteinase-activated receptors (PAR-2 and PAR-1, respectively). The therapeutic hypothesis is that PPC-001 targets and eradicates cancer stem cells, reverses epithelial-to-mesenchymal transition (EMT) to restore an epithelial phenotype, inhibits tumor angiogenesis, and remodels the fibrotic tumor microenvironment, thereby preventing tumor recurrence and metastasis without the severe toxicities of traditional chemotherapy.
