PPI-1011 is a synthetic plasmalogen precursor designed to augment plasmalogen levels in patients with disorders of plasmalogen deficiency, most notably Rhizomelic chondrodysplasia punctata (RCDP), an ultra-rare genetic disorder characterized by significant physical and cognitive delays due to low circulating plasmalogens. The drug is orally administered and has demonstrated the ability to significantly increase serum levels of target plasmalogens in both single and multiple ascending dose studies. Mechanistically, PPI-1011 acts as a prodrug that delivers ether lipid precursors for endogenous biosynthesis of functional plasmalogens throughout the body, including the brain. Preclinical data also suggest neuroprotective effects potentially related to its delivery of docosahexaenoic acid (DHA) and lipoic acid moieties, which may contribute anti-apoptotic and antioxidant properties.
