Overview

PPI-383 is a selective, potent non-nucleoside inhibitor of the hepatitis C virus (HCV) NS5B RNA-dependent RNA polymerase. It demonstrates pan-genotypic antiviral activity against major HCV genotypes (1a, 1b, 2a, 3a, and 4a) with low nanomolar EC50 values in replicon assays. The compound binds to the Palm II pocket of NS5B and exhibits favorable pharmacokinetic properties including good oral bioavailability and stability in preclinical species. PPI-383 was developed by Presidio Pharmaceuticals as a potential oral therapy for chronic hepatitis C infection and has been evaluated in early-phase clinical trials for safety, pharmacokinetics, and antiviral efficacy in both healthy volunteers and HCV-infected patients[1][2][3][4][5].
