Overview

PPI-801 (also known as PLX-801) is a potent and selective small molecule inhibitor of p38 alpha mitogen-activated protein (MAP) kinase (MAPK14). Developed by Plexxikon using its proprietary scaffold-based drug discovery platform, the compound was designed to treat inflammatory conditions by blocking the production of pro-inflammatory cytokines, including tumor necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β). PPI-801 demonstrated significant efficacy in preclinical models of rheumatoid arthritis and other inflammatory diseases. However, its clinical development was discontinued in the mid-2000s as Plexxikon shifted its focus toward other kinase inhibitors and multi-kinase inhibitors like PLX-647. Note that the code name PPI-801 has also been used in academic literature to describe a synthetic DHA-plasmalogen precursor developed by PhenoMene (now Prodrome Sciences) for neurodegenerative research.
