Drug intelligence / Profile preview
PPI-Fc is a recombinant fusion protein consisting of the type 1 diabetes autoantigen preproinsulin (PPI) fused to the Fc fragment of an immunoglobulin G (IgG) antibody. Developed by researchers at the Institut Cochin, INSERM, and partner French institutions, PPI-Fc is designed to prevent type 1 diabetes by inducing central immune tolerance. The Fc portion of the fusion protein binds to the neonatal Fc receptor (FcRn), enabling its active transport across the placenta from pregnant mothers to fetuses, or across the intestinal epithelium in neonates. Once transferred, PPI-Fc is taken up by migratory dendritic cells and ferried to the thymus, where it promotes the deletion of autoreactive T cells and the generation of preproinsulin-specific regulatory T cells (Tregs), thereby halting the autoimmune cascade that leads to beta-cell destruction.
