Overview

pPJV2012 is a DNA-encoded immunostimulator consisting of a plasmid that expresses human Interleukin-12 (IL-12). Developed by PowderJect Pharmaceuticals (later acquired by Chiron and subsequently Novartis), it was designed as a genetic adjuvant to enhance the cellular immune response to DNA vaccines. The plasmid is typically administered via Particle Mediated Epidermal Delivery (PMED), a "gene gun" technology that delivers DNA-coated gold microparticles directly into the epidermis. In clinical development, pPJV2012 was evaluated in combination with pPJV7630, a therapeutic DNA vaccine targeting Herpes Simplex Virus type 2 (HSV-2), to boost Th1-biased T-cell responses in latently infected individuals. Following the acquisition of Chiron by Novartis, the program was discontinued.
