PPM1D siRNA is a small interfering RNA molecule designed to target and reduce the expression of the PPM1D gene (encoding protein phosphatase, Mg2+/Mn2+ dependent, 1D). PPM1D acts as an oncogene in various cancers, including pediatric gliomas such as diffuse intrinsic pontine glioma (DIPG), often through truncation mutations leading to increased enzymatic stability and phosphatase activity. By silencing PPM1D using siRNA, proliferation of tumor cells carrying PPM1D mutations is inhibited, at least in part through reactivation of cell cycle checkpoints and pro-apoptotic pathways involving proteins such as p53 and Chk1. PPM1D siRNA has been tested as a monotherapy and in combination with panobinostat in preclinical models of DIPG, resulting in reduced tumor cell viability and enhanced sensitivity to chemotherapeutic agents. The primary modality is as a gene-silencing therapy for targeted treatment of PPM1D-mutated cancers, especially brain tumors such as DIPG[3][10].
