**PpPfs25** is a Pichia-expressed recombinant form of the *Plasmodium falciparum* sexual-stage surface antigen Pfs25. It is a protein-subunit transmission-blocking malaria vaccine antigen intended to elicit anti-Pfs25 antibodies; when a mosquito ingests these antibodies in a blood meal, they inhibit parasite development in the mosquito and thereby reduce onward transmission. PpPfs25 was evaluated by NIAID in a Phase 1 study as the adjuvanted formulation PpPfs25/ISA51. The trial showed induction of Pfs25-specific, transmission-blocking antibodies, but the ISA51 formulation caused substantial local reactogenicity and systemic adverse events, limiting further development of that formulation.
