Drug intelligence / Profile preview
PPSK (polydopamine-polyethylenimine-l-serine-Klotho plasmid nanoparticles) is a kidney-targeted non-viral gene therapy designed to prevent the transition of acute kidney injury (AKI) to chronic kidney disease (CKD) and renal fibrosis. Developed by researchers at Sun Yat-sen University and Jinan University, PPSK consists of a plasmid encoding the anti-aging protein Klotho, encapsulated within polydopamine-polyethylenimine-l-serine nanoparticles. The nanoparticles selectively target injured renal tubular epithelial cells (TECs) by binding to kidney injury molecule-1 (KIM-1). Upon delivery, PPSK restores and maintains Klotho protein expression, which upregulates peroxisome proliferator-activated receptor alpha (PPARα) by inhibiting p38 and JNK phosphorylation. This cascade improves tubular fatty acid beta-oxidation and reduces renal lipid accumulation, ultimately protecting against kidney fibrosis.
