Drug intelligence / Profile preview
PR-364 is a selective, small-molecule activator of the E3 ubiquitin ligase Parkin (encoded by the PRKN/PARK2 gene), developed by Progenra. Discovered via high-throughput screening, PR-364 selectively promotes Parkin-dependent mitophagy and mitochondrial biogenesis without affecting other closely related E3 ligases. By enhancing the clearance of damaged mitochondria and boosting mitochondrial function, PR-364 has demonstrated therapeutic potential in preclinical models of acute pancreatitis, myocardial infarction (protecting against mortality and preventing progression to heart failure), and other conditions characterized by mitochondrial dysfunction.
