PR1 peptide is a **9-amino acid HLA-A2-restricted peptide vaccine** derived from the myeloid proteins proteinase 3 and neutrophil elastase. It is administered as a cancer vaccine and investigated primarily for the treatment of **acute myeloid leukemia (AML)**, **chronic myeloid leukemia (CML)**, and **myelodysplastic syndrome (MDS)**. The peptide is recognized on leukemic cells by cytotoxic T lymphocytes (CTLs), which preferentially kill these malignant cells. PR1 vaccination stimulates a CTL immune response targeted at cells displaying PR1 peptide in the context of HLA-A2, resulting in **tumor cell lysis**. Clinical trials have shown PR1 peptide vaccination to be **immunogenic and associated with objective clinical responses, including molecular remission**, in patients with AML, CML, or MDS. The vaccine is administered subcutaneously, often formulated with Montanide ISA51 and sometimes co-administered with GM-CSF as an adjuvant[1][2][3][5][7][9].
