Overview

PR1-specific T lymphocytes are an adoptive cellular immunotherapy developed at M.D. Anderson Cancer Center for the treatment of chronic myelogenous leukemia (CML), particularly in patients with recurrent or persistent disease following allogeneic hematopoietic transplantation. The therapy consists of donor-derived cytotoxic T lymphocytes (CTLs) that are expanded ex vivo and primed to recognize PR1, an HLA-A2-restricted nonapeptide derived from the leukemia-associated antigens proteinase 3 and neutrophil elastase. By targeting the PR1 peptide presented on HLA-A2 molecules on the surface of leukemia cells, these CTLs selectively induce cell death in malignant cells while sparing normal hematopoietic tissues, thereby enhancing the graft-versus-leukemia effect without the associated risk of graft-versus-host disease.
