PR104 + filgrastim is a combination therapy consisting of **PR104**, a hypoxia-activated prodrug, and **filgrastim**, a recombinant granulocyte colony-stimulating factor. PR104 is a phosphate ester that is rapidly converted to PR-104A, which is then enzymatically reduced to cytotoxic nitrogen mustards under hypoxic conditions, causing DNA cross-linking and cell death, especially in hypoxic tumor microenvironments[1]. Filgrastim stimulates the production of neutrophils to mitigate the myelosuppressive effects of cytotoxic chemotherapy, such as PR104, reducing risks associated with neutropenia[2][4][5]. This combination has been investigated to determine the maximum tolerated dose and safety in patients with solid tumors[2][4][5].
