PR201 is an innovative antibody-cytokine fusion protein developed by Shandong Boan Biotechnology Co., Ltd. It consists of a high-affinity monoclonal antibody targeting programmed cell death protein 1 (PD-1) fused to a masked interleukin-2 (IL-2) cytokine. The drug is designed as a symmetric prodrug; the IL-2 component remains inactive in systemic circulation to minimize toxicities such as vascular leak syndrome. Upon localization to the tumor microenvironment via the anti-PD-1 domain, the IL-2 is selectively activated through enzymatic cleavage and cis-activation. This dual-action approach combines immune checkpoint blockade with localized T-cell and natural killer (NK) cell stimulation. PR201 is being investigated for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and melanoma.
