Drug intelligence / Profile preview
PR3mutB-CATCR-T cells is an investigational chimeric autoantigen T cell receptor (CATCR)-T cell therapy being developed for the treatment of proteinase 3 (PR3)-specific antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) [0928]. CATCR-T cells express a disease-relevant autoantigen fused to a modified T cell receptor, enabling selective targeting of autoreactive B cells through their B cell receptors [0928]. To avoid off-target binding to physiologic ligands such as NB1 (CD177) on normal cells, PR3mutB-CATCR-T cells utilize a structure-guided engineered PR3 variant (PR3mutB) that eliminates off-target binding while maintaining potent cytotoxicity against pathogenic PR3-ANCA+ B cells [0928].
