PR55α RNAi refers to an experimental RNA interference-based therapeutic approach designed to silence the expression of PR55α, a regulatory subunit of the Protein Phosphatase 2A (PP2A) holoenzyme. Research, primarily conducted at the University of Nebraska Medical Center, has identified PR55α as a key promoter of pancreatic tumorigenesis, where it is significantly overexpressed and correlates with poor patient survival. By utilizing RNAi to deplete PR55α levels, this strategy aims to inhibit multiple oncogenic signaling pathways, including AKT, ERK1/2, and Wnt/β-catenin. In preclinical models of pancreatic ductal adenocarcinoma, PR55α depletion has been shown to significantly impair cell growth, clonogenicity, and mobility, while reducing tumorigenicity and distant metastasis in vivo.
