PR61 is a preclinical mRNA-based therapeutic developed by TheraPTEN designed to restore functional PTEN activity in PTEN-deficient cancer cells. The therapy consists of mRNA encoding a patented, secretable, and cell-permeable albumin-PTEN fusion protein encapsulated within a lipid nanoparticle (LNP). Following intravenous administration, the mRNA is primarily taken up and expressed by the liver, which then secretes the functional PTEN fusion protein into the systemic circulation. This "Trojan Horse" delivery mechanism allows the protein to enter distant tumor cells, where it acts as a tumor suppressor by antagonizing the PI3K/AKT/mTOR signaling pathway. PR61 is being investigated for orphan cancer indications, major PTEN-deficient cancers, and potentially for longevity and healthy aging applications.
