PR68 is a novel albumin–PTEN fusion protein designed to restore tumor suppressor function in cancers where PTEN is inactivated. Developed by TheraPTEN in collaboration with 4basebio, PR68 is engineered to be secreted and targeted to cancer cells via albumin-mediated uptake. It utilizes a novel receptor for cell permeability, allowing the PTEN moiety to enter the cytoplasm and activate cell death pathways. In the context of 4basebio's platform, PR68 is delivered as a DNA-encoded therapeutic using synthetic linear hairpin DNA (hpDNA) encapsulated within Hermes lipid nanoparticles (LNPs), offering a thermostable and durable alternative to mRNA-based biologics.
