PR8 deltaNS1

PR8 deltaNS1 is a laboratory-engineered, live attenuated influenza A/Puerto Rico/8/34 (H1N1) virus in which the nonstructural protein 1 (NS1) gene is deleted, abolishing a key viral interferon antagonist and thereby strongly attenuating replication in interferon-competent hosts while enhancing innate and adaptive immune activation.[1][3][5] Deletion of NS1 removes multiple functions that normally suppress RIG-I–mediated signaling, PKR activation, and type I interferon and inflammatory cytokine production, resulting in robust induction of antiviral genes, dendritic cell activation, and T-cell and antibody responses.[1][3][5] PR8 deltaNS1 has been developed and widely used as a preclinical live attenuated influenza vaccine platform and as an immunogenic vector to study mucosal and systemic immunity, with protective efficacy against homologous and heterosubtypic influenza virus challenge in animal models.[3][5]
