Overview

PRA-523 is a first-in-class topical small molecule antagonist of Protease-Activated Receptor 2 (PAR2), currently being developed by Praeventix for the treatment of pruritus associated with atopic dermatitis. PAR2 is a G-protein coupled receptor that is highly expressed in the skin and plays a pivotal role in mediating itch and neurogenic inflammation. By selectively inhibiting PAR2, PRA-523 aims to block the signaling pathways that trigger the sensation of itch and the subsequent inflammatory response, thereby breaking the itch-scratch cycle. The drug is formulated for topical administration and has been evaluated in Phase I/II clinical trials in New Zealand to determine its safety, pharmacokinetics, and efficacy in reducing pruritus in patients with atopic dermatitis.
