DS-6016a (prafnosbart) is a humanized IgG1-kappa monoclonal antibody that targets the activin A receptor type 1 (ACVR1, also known as ALK2). It was developed by Daiichi Sankyo and Saitama Medical University for the treatment of fibrodysplasia ossificans progressiva (FOP), a rare genetic disorder characterized by progressive heterotopic ossification. In FOP, mutations in ACVR1/ALK2 lead to abnormal bone formation due to hyper-responsiveness to bone morphogenetic proteins (BMPs) and aberrant signaling via Activin A. By inhibiting ALK2, DS-6016a aims to block this pathological signaling pathway and prevent disease progression. The drug has been evaluated in phase 1 clinical trials for safety, tolerability, and pharmacokinetics in healthy volunteers[1][5][7].
