A therapeutic regimen combining pralatrexate (a folate analog metabolic inhibitor) with vitamin B12 and folic acid supplementation. This combination is designed to allow for effective cancer treatment while mitigating toxicity, particularly mucositis. Pralatrexate competitively inhibits dihydrofolate reductase (DHFR) and selectively enters cells expressing reduced folate carrier type 1 (RFC-1), which is overexpressed on certain cancer cells. The vitamin supplementation is specifically added to reduce treatment-related toxicities.
