Praluzatamab ravtansine is an investigational **conditionally activated Probody drug conjugate** targeting activated leukocyte cell adhesion molecule CD166. The single molecular entity comprises a masked recombinant anti-CD166 monoclonal antibody conjugated to the maytansinoid microtubule-disrupting payload DM4. Tumor-associated proteases remove the peptide mask preferentially in the tumor microenvironment, enabling CD166 binding, internalization, and intracellular DM4 delivery while aiming to limit normal-tissue target engagement. CytomX developed CX-2009 with ImmunoGen payload-conjugation technology; the agent was evaluated in Phase I/II studies in advanced solid tumors and Phase II breast-cancer studies. CytomX stated in December 2022 that it had elected not to advance the program further without a partner.
