Overview

PRAME SPEAR T-cell is an autologous T cell therapy genetically engineered to express a high-affinity T cell receptor (TCR) specific for the cancer-testis antigen **PRAME** (Preferentially Expressed Antigen in Melanoma). The therapy is designed to recognize and kill tumor cells presenting PRAME peptides in the context of HLA-A*02. PRAME is highly expressed in various solid tumors (including melanoma, sarcoma, ovarian, and triple-negative breast cancers), as well as certain hematologic cancers, but has limited expression in normal tissues, making it a promising immunotherapy target. The therapy uses "SPEAR" (Specific Peptide Enhanced Affinity Receptor) technology to enhance the TCR's ability to recognize tumor-associated PRAME. The primary indications under investigation include advanced or refractory solid tumors expressing PRAME, with ongoing development in other cancer types. The product is manufactured by engineering the patient’s own T cells ex vivo, expanding them, and then reinfusing them after lymphodepletion. The drug is being developed and/or trialed by Adaptimmune, and was previously associated with a collaboration with GSK[8].
