PRAME (Preferentially Expressed Antigen in Melanoma) targeting therapies encompass two primary modalities: engineered T-cell receptor (TCR) T-cell therapies and recombinant protein vaccines. The TCR-T approach, developed using Adaptimmune's SPEAR (Specific Peptide Enhanced Affinity Receptor) platform, involves engineering a patient's T-cells to express high-affinity receptors specific to PRAME peptides presented by HLA molecules, enabling the targeted destruction of PRAME-positive tumor cells. This program was a key component of a strategic collaboration between Adaptimmune and GSK, which was terminated in late 2022, with rights reverting to Adaptimmune in 2023. Separately, GSK developed a PRAME immunotherapeutic consisting of a recombinant PRAME protein combined with the AS15 immunostimulant. This vaccine was evaluated in Phase I trials for surgically resected non-small cell lung cancer (NSCLC) to induce humoral and cellular immune responses, though further development was halted following disappointing results with similar immunotherapeutics in the same indication.
