PRAME-targeted TCR-engineered iNKT cell therapy (CF-033) is an allogeneic, off-the-shelf cell-based biologic therapy being developed by MiNK Therapeutics in collaboration with the C-Further international pediatric oncology consortium. The therapy consists of donor-derived invariant natural killer T (iNKT) cells that have been genetically engineered to express a T cell receptor (TCR) specific for the tumor-associated antigen PRAME (Preferentially Expressed Antigen in Melanoma). This approach leverages the unique properties of iNKT cells, which possess both innate and adaptive immune functions and can home to tumor tissues without the requirement for HLA matching. CF-033 is designed to treat a variety of PRAME-expressing malignancies, including pediatric sarcomas, acute myeloid leukemia, and medulloblastoma, as well as adult solid tumors such as non-small cell lung cancer (NSCLC), ovarian cancer, and melanoma. The program is currently in preclinical development, with funding secured for IND-enabling studies as of early 2026.
