PRAME TCR T cells are a type of adoptive cellular immunotherapy where T cells are genetically engineered to express a T-cell receptor (TCR) that specifically recognizes the Preferentially Expressed Antigen in Melanoma (PRAME). PRAME is a cancer-testis antigen that is highly expressed in a wide range of solid tumors (such as melanoma, uveal melanoma, and synovial sarcoma) and hematological malignancies (such as multiple myeloma and acute myeloid leukemia), while maintaining restricted expression in healthy tissues. These engineered T cells identify PRAME-derived peptides presented by Human Leukocyte Antigen (HLA) complexes, most commonly HLA-A*02:01. Upon recognition of the peptide-MHC complex, the TCR-T cells undergo activation, proliferation, and direct lysis of the tumor cells. Advanced iterations of this therapy, such as Bellicum's Go-TCR platform, incorporate inducible co-stimulatory switches (e.g., MyD88/CD40) to enhance T-cell persistence, expansion, and efficacy within the immunosuppressive tumor microenvironment.
