**Pranidipine** is a long-acting, 1,4-dihydropyridine calcium channel blocker with antihypertensive activity. It acts mainly by inhibiting L-type calcium channels in vascular smooth muscle, leading to vasodilation and a reduction in blood pressure. It is classified as a third-generation dihydropyridine derivative, distinguished by a prolonged duration of action. Pranidipine can be administered once daily and has demonstrated dose-dependent blood pressure-lowering effects without clinically significant changes in heart rate. The mechanism of action is closely related to that of other dihydropyridine calcium channel blockers, targeting voltage-dependent calcium influx in vascular smooth muscle. Pranidipine may also enhance acetylcholine-induced and nitric oxide-dependent vasorelaxation by mechanisms not mediated by cyclic GMP.
