Overview

Prasinezumab is a humanized IgG1 monoclonal antibody that selectively binds to aggregated α-synuclein. By targeting these aggregates—which are implicated in the pathogenesis of Parkinson's disease and other α-synucleinopathies such as dementia with Lewy bodies—prasinezumab aims to reduce neuronal toxicity and potentially slow disease progression. The drug is under development primarily for early-stage Parkinson's disease. Preclinical studies showed reduction in neurotoxic forms of α-synuclein and decreased propagation between cells. Clinical trials have demonstrated that while primary endpoints were not met in some studies (such as overall MDS-UPDRS scores), there was evidence suggesting slower motor progression in certain subgroups of patients with rapidly progressing Parkinson’s disease. Prasinezumab continues to be evaluated for its potential as a disease-modifying therapy[1][2][4][5][6][7][8].
