Drug intelligence / Profile preview
PRC1584 (also known as MMV1803522) is a novel, orally bioavailable antimalarial drug candidate belonging to the β-carboline carboxamide class. Developed by researchers at Virginia Tech, the University of Georgia, and collaborating institutions, PRC1584 was identified as a potent lead compound from a virtual ligand screen based on the tetrahydro-β-carboline MMV008138. Unlike MMV008138, which targets the MEP pathway, PRC1584 operates via a distinct, novel mechanism of action that does not involve MEP pathway inhibition. It exhibits nanomolar potency against the asexual intraerythrocytic stages of *Plasmodium falciparum*, including both actively proliferating ring stages and dihydroartemisinin (DHA)-induced dormant/quiescent stages, regardless of artemisinin resistance. PRC1584 has demonstrated a high barrier to resistance in vitro and in vivo efficacy in *Plasmodium berghei*-infected mouse models.
