Overview

**Precemtabart tocentecan** is an investigational, CEACAM5-directed antibody-drug conjugate developed by Merck KGaA for advanced solid tumors, led by metastatic colorectal cancer. The ADC binds carcinoembryonic antigen-related cell adhesion molecule 5 on tumor cells and delivers an exatecan topoisomerase I inhibitor payload through a circulation-stable beta-glucuronide linker, causing topoisomerase I inhibition and DNA damage after tumor-cell uptake. It is being evaluated in the Phase 3 PROCEADE-CRC-03 trial in metastatic colorectal cancer, as well as earlier-stage studies in other CEACAM5-expressing solid tumors.
