Prednisone + belimumab is a combination therapy used primarily in the management of systemic lupus erythematosus (SLE) and lupus nephritis. Prednisone is a synthetic glucocorticoid corticosteroid that exerts broad anti-inflammatory and immunosuppressive effects by modulating gene expression, reducing cytokine production, and suppressing immune cell activity. Belimumab is a fully human monoclonal antibody targeting B-lymphocyte stimulator protein (BLyS/BAFF), thereby inhibiting B cell survival and differentiation. This combination is not available as a fixed-dose product but represents the concurrent use of both agents as part of standard-of-care regimens for SLE, often with other immunosuppressants or antimalarials. The addition of belimumab to prednisone-containing regimens has been shown to improve disease control, reduce disease activity scores, and enable more rapid reduction in glucocorticoid doses while maintaining efficacy[1][6][7].
