Chemotherapy Regimen 1 is a multi-agent chemotherapy protocol utilized in the Phase 1 AUGMENT-102 clinical trial (NCT05360160) sponsored by Syndax Pharmaceuticals. This regimen is evaluated in combination with the menin-KMT2A inhibitor revumenib (SNDX-5613) for the treatment of pediatric and adult patients with relapsed or refractory acute leukemias, including acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), mixed phenotype acute leukemia (MPAL), and acute undifferentiated leukemia (AUL), specifically those harboring KMT2A rearrangements, NPM1 mutations, or NUP98 alterations. The regimen is administered in two distinct cycles: Cycle 1 consists of prednisone, vincristine, pegaspargase (or calaspargase pegol), and daunorubicin; Cycle 2 consists of etoposide and cyclophosphamide. This combination leverages multiple cytotoxic mechanisms, including DNA alkylation, topoisomerase inhibition, tubulin disruption, and asparagine depletion, to achieve cytoreduction in aggressive leukemic subtypes.
