Overview

Pregabalin + celecoxib is a fixed-dose combination of two small molecule drugs used primarily for the management of pain, particularly in conditions involving both neuropathic and nociceptive mechanisms such as chronic low-back pain, acute postoperative pain, and lumbar disc herniation. Pregabalin is an anticonvulsant that binds to the alpha2-delta subunit of presynaptic voltage-gated calcium channels in the central nervous system, thereby reducing the release of excitatory neurotransmitters like glutamate, norepinephrine, substance P, and dopamine. Celecoxib is a selective cyclooxygenase-2 (COX-2) inhibitor that reduces inflammation and pain by inhibiting prostaglandin synthesis. The combination has demonstrated superior analgesic efficacy compared to either drug alone and can reduce opioid requirements postoperatively with similar rates of adverse effects as monotherapy[1][2][4][7].
