Overview

The combination of pregabalin and imipramine is used for treating painful polyneuropathy. This combination therapy uses moderate doses of both medications to provide better pain relief than monotherapy with either drug alone. Pregabalin is an anticonvulsant that binds to the α2δ subunit of voltage-dependent calcium channels, while imipramine is a tricyclic antidepressant that inhibits serotonin and norepinephrine reuptake. Clinical trials have shown that the combination provides significantly better pain reduction compared to either medication used alone, though it may cause more side effects including dizziness, drowsiness, confusion, and difficulty concentrating.
