pregabalin + oxaliplatin

Pregabalin + oxaliplatin (PreOx) refers to a clinical investigation of the co-administration of pregabalin, a small molecule alpha-2-delta calcium channel ligand, alongside oxaliplatin-based chemotherapy. Pregabalin is primarily used to treat neuropathic pain by binding to the alpha-2-delta subunit of voltage-gated calcium channels, thereby reducing the release of excitatory neurotransmitters. Oxaliplatin is a platinum-based antineoplastic agent that acts as a DNA alkylator to inhibit DNA synthesis in cancer cells. The combination was evaluated in the Phase 3 PreOx trial, led by the University of São Paulo with support from Pfizer, to determine if preemptive pregabalin could prevent or reduce oxaliplatin-induced painful peripheral neuropathy (OXAIPN) in patients with colorectal cancer. The trial ultimately demonstrated that pregabalin did not significantly decrease the incidence of chronic oxaliplatin-related neuropathic pain compared to placebo.
