Drug intelligence / Profile preview
Pregabalin naproxencarbil (NG1706, XG005) is an oral, non-opioid, small-molecule dual-acting prodrug conjugate of the NSAID naproxen and the gabapentinoid pregabalin. Originally developed by Xgene Pharmaceutical and licensed to NeuroGen Pharma in Greater China, the drug is designed to treat both inflammatory and neuropathic pain. Upon absorption, it is metabolized into naproxen and pregabalin, which simultaneously inhibit cyclooxygenase (COX) enzymes and modulate the alpha-2/delta-1 subunit of voltage-gated calcium channels (CACNA2D1). This dual mechanism of action targets multiple pain pathways to provide broad-spectrum analgesia with improved gastrointestinal and central nervous system safety compared to traditional therapies. Pregabalin naproxencarbil has completed Phase IIb/III clinical trials for acute post-surgical pain following bunionectomy in the US and Phase IIb trials for chronic osteoarthritis pain in China.
