Pregnenolone acetate is a synthetic pregnane steroid and an ester derivative of pregnenolone. It has been described as a glucocorticoid and is used primarily as a skin-conditioning and anti-aging agent in cosmetic formulations[8]. Historically, it was investigated for hormone replacement therapy, neuroprotection (notably in neurodegenerative diseases such as Alzheimer's disease), stress and anxiety management, anti-inflammatory applications (such as arthritis), and skin health enhancement[5][8]. Mechanistically, it acts as a neurosteroid with potential modulatory effects on neuronal function. Pregnenolone itself is known to be a negative allosteric modulator of the CB1 receptor, an agonist of the pregnane X receptor, and interacts with microtubule-associated proteins; these activities may also be relevant for its ester derivatives like pregnenolone acetate[1][5].
