Pretargeted glycoengineered NK cell (PG-NK) is a first-in-class cell therapy platform designed to overcome limitations of traditional Chimeric Antigen Receptor T cell (CAR-T) therapy, such as complex manufacturing, high cost, toxicity, and resistance. This platform intentionally decouples the targeting moiety (ligand) from the effector cell (Natural Killer cell) using bioorthogonal click chemistry. It involves universally glycoengineered NK cells (NK-TZ) that are paired with tumor-specific antibodies functionalized with complementary click partners (Ab-TCO). This allows for rapid, antigen-flexible targeting without genetic modification of the NK cells. The platform has demonstrated enhanced cytotoxicity and tumor elimination in preclinical models of lymphoma and breast cancer, with an 'off-the-shelf' potential.
