Overview

Prezalumab (AMG 557) is a fully human IgG2 monoclonal antibody developed for the treatment of autoimmune diseases. It specifically targets and inhibits the function of inducible T-cell co-stimulator ligand (ICOSL), also known as B7-related protein 1 (B7RP-1), thereby modulating T cell activation and immune responses. The drug was primarily investigated for systemic lupus erythematosus (SLE), Sjogren's syndrome, cutaneous lupus erythematosus, and psoriasis. Prezalumab was developed by Amgen in partnership with MedImmune/AstraZeneca but has been discontinued in all indications[4][6][3][5]. Its mechanism involves blocking the interaction between ICOSL on antigen-presenting cells and ICOS on activated T cells to reduce pathological immune activation[6][2].
